UCHL3 is a deubiquitinating enzyme (DUB) that controls levels of cellular ubiquitin through processing of ubiquitin precursors and ubiquitinated proteins. It is a thiol protease that recognizes and hydrolyzes a peptide bond at the C-terminal glycine of either ubiquitin or NEDD8. It digests precursors and ubiquitinated proteins to generate monomeric ubiquitin. This UCHL3 is recombinatly expressed in E.coli.

The severe acute respiratory syndrome coronavirus papain-like protease (SARS-CoV His6-PLpro) is involved in the processing of the viral polyprotein. Proteolytic processing of the coronavirus replicase poly-protein is essential for generating a functional virus replication complex. PLpro is able to hydrolyze both ISG15-Rhodamine110 or diubiquitin/tetra-ubiquitin substrates, but is very inefficient when processing mono-Ub conjugates or synthetic peptide substrates.
